Des chercheurs ont rajeuni des souris en activant quatre gènes : les facteurs Yamanaka. Les effets étaient visibles aussi bien sur l'organisme entier qu'à l'intérieur des cellules. Les facteurs Yamanaka sont quatre gènes qui, s'ils sont activés dans des cellules en culture, peuvent les faire revenir à un état embryonnaire. Ces cellules pluripotentes induites sont capables de fournir toutes les cellules de l'organisme et de se diviser à l'infini. D'où l'idée d'activer ces facteurs pour faire rajeunir un organisme. L'ennui toutefois est que, comme l'ont montré des études précédentes, l'induction de ces gènes chez des animaux provoque des cancers. Pour éviter ce problème, des chercheurs du Salk Institute for Biological Studies (la Jolla) ont choisi d'induire les facteurs Yamanaka sur de courtes périodes répétées. Tout d'abord, ils ont travaillé sur des souris qui avaient la progéria, une maladie humaine qui fait vieillir anormalement vite. Une équipe de biologistes de l'Inserm a rendu leur jeunesse à des cellules humaines prélevées chez des personnes âgées et atteintes de sénescence, le vieillissement cellulaire serait donc réversible. Jean-Marc Lemaître, qui a mené cette étude, explique à Futura-Sciences les clés de cette étonnante découverte. Jusqu'à présent, les biologistes savaient rendre leur pluripotence , c'est-à-dire la capacité à se différencier, à des cellules qui étaient déjà différenciées (devenues donc des cellules d'un certain type, peau, muscles...). Mais grâce aux travaux d'une équipe de l'Inserm, menée par Jean-Marc Lemaître de l'université de Montpellier, la science a fait un nouveau pas en avant : il est possible de redonner leur pluripotence à des cellules âgées, ainsi que leur jeunesse ! Le processus de vieillissement est donc réversible. Tout a commencé en 2007, quand le Japonais Shinya Yamanaka et ses collègues mettent au point un cocktail éponyme de 4 gènes (Oct3/Oct4, SOX2, Klf4 et c-myc) qui, incorporé dans le génome d'une cellule différenciée grâce à un rétrovirus, la fait revenir au stade de cellule souche pluripotente (on parle de cellules pluripotentes induites). Un pas énorme pour la médecine régénérative.

Des cellules pluripotentes mais toujours vieilles

Mais la technique se heurte à un problème, comme l'explique Jean-Marc Lemaître à Futura-Sciences : « Quatre études ont montré que la sénescence était une barrière à cette reprogrammation ». En d'autres termes, il est possible de redonner sa pluripotence à une cellule différenciée, à condition qu'elle ne soit pas atteinte de sénescence. Or parmi les cas nécessitant un traitement via la médecine régénérative, certains sont des patients âgés ou possédant des cellules qui vieillissent prématurément.
Qu'est-ce qu'une cellule sénescente ? C'est une cellule qui ne prolifère plus. Lorsqu'une cellule subit un stress, elle finit par entrer dans un état de sénescence ou, si le stress est important, déclenche l'apoptose (mort cellulaire). « Nous avons travaillé sur des cellules de patients âgés altérées par le vieillissement que nous avons en plus conduites en sénescence, détaille Jean-Marc Lemaître. Les cellules âgées ont un métabolisme altéré, tout comme l'expression de leurs gènes. »

Un nouveau cocktail à 6 gènes pour déprogrammer une cellule

Pour cela, les scientifiques ont mis en culture des cellules de patients âgés, plus que centenaires pour certains, déjà altérées par le vieillissement, jusqu'à ce qu'elles deviennent incapables de proliférer. C'est sur ces cellules, ainsi que sur d'autres pas encore altérées car appartenant à des patients moins âgés, qu'ils ont effectué leurs tests. Ils ont ajouté 2 gènes au cocktail Yamakana (les gènes Nanog et Lin28). « Vingt jours environ après l'injection du cocktail dans les cellules, nous avons observé une reprolifération des cellules, puis des colonies de cellules pluripotentes après 3 à 4 semaines. » Les biologistes étaient parvenus non seulement à rendre leur pluripotence à ces cellules sénescentes et différenciées, mais également leur jeunesse !

Le vieillissement cellulaire est réversible

« Après cela, nous avons effectué une série d'expériences afin de vérifier que toutes les marques de sénescence et du vieillissement avaient bien été effacées », précise Jean-Marc Lemaître. Il a fallu notamment vérifier l'état des télomères, fragments d'ADN situés à l'extrémité des chromosomes et dont la longueur est un très bon indicateur de la capacité de prolifération des cellules. Après le traitement de jouvence, les télomères avaient retrouvé une longueur conférant aux cellules une capacité de prolifération accrue. « Nous avons également vérifié la physiologie de la cellule, complète le biologiste. L'activité mitochondriale, le profil d'expression génique... » Tout indiquait que la cellule avait retrouvé sa jeunesse. « Nous avons démontré la réversibilité du vieillissement cellulaire », conclut Jean-Marc Lemaître avec enthousiasme. La prochaine étape est de mettre ce protocole en application et de s'assurer qu'il ne présentera aucune nocivité pour les patients. Ces travaux, à paraître dans la revue Genes & Development, représentent un réel espoir pour traiter des patients dont les cellules ou les tissus sont sujets à un vieillissement en général et dans des cas pathologiques, à un vieillissement précoce.
